铜死亡是一种铜(Cu)依赖的非凋亡型调节性细胞死亡(RCD)方式,其细胞学机制主要是超载的铜离子靶向线粒体中三羧酸(TCA)循环的硫辛酰化蛋白,导致铁硫簇蛋白丢失,进而引发细胞蛋白毒性应激. 铜死亡已经被证明与包括癌症在内的多种疾病密切相关. 自2022年其机制被阐明后,利用纳米材料介导的铜死亡效应来治疗疾病的相关研究也成为新的研究热点. 文章阐述了细胞内铜代谢的过程与铜死亡的分子机制,重点介绍了目前基于纳米材料诱导的铜死亡作为癌症治疗手段的研究进展,并对该领域面临的挑战和未来的发展方向进行讨论. |

Research progress on nanomaterial-induced cuproptosis for cancer therapy |

Cuproptosis is a copper(Cu)-dependent non-apoptotic regulatory cell death(RCD) mode, and its cellular mechanism is that thioacylated proteins of the tricarboxylic acid(TCA) cycle in overloaded copper ion targeting mitochondria lead to the loss of iron-sulfur cluster proteins and subsequently cellular protein toxicity stress. Cuproptosis has been proven to be closely associated with various diseases, including cancer. Since its mechanism was elucidated in 2022, researches related to the use of nanomaterial-mediated cuproptosis effects to treat diseases have become a new research hotspot. This article describes the process of intracellular copper metabolism and the molecular mechanism of cuproptosis, highlights the current research progress based on nanomaterial-induced cuproptosis as a cancer therapeutic tool, and discusses the challenges and future directions in this field. |
