菠菜YABBY基因家族鉴定与表达分析

摘要:
YABBY基因家族在植物叶片发育中起着关键作用,为克隆和研究菠菜YABBY基因家族成员基因及其生物学功能,本研究通过生物信息学方法鉴定出4个菠菜YABBY基因家族成员,依次命名为SoYAB1~4;亚细胞定位结果表明3个基因定位在细胞核,1个定位在细胞壁;保守性分析菠菜YABBY基因家族由C2C2锌指结构域与YABBY结构域组成,成员之间相对保守。系统发育关系结果将4个序列分成4个亚族;启动子和表达分析表明,菠菜YABBY基因主要在叶片中表达,尤其在功能性叶片中表达最为显著。菠菜YABBY基因家族成员的表达受低温、盐胁迫和多种激素胁迫的影响。这些研究结果为后期进行YABBY基因的克隆与功能分析提供理论基础,为理解菠菜叶片发育的分子机制提供了理论支撑。

Identification and Expression Analysis of YABBY Gene Family in Spinach

Abstract:
The YABBY gene family plays a key role in plant leaf development. In order to clone and study the function of spinach YABBY gene family members, in this study, we identified four spinach YABBY gene family members by bioinformatics methods, named SoYAB1~4; the results of the subcellular localization showed that three genes were localized in the nucleus and one in the cell wall; Conservative analysis of spinach YABBY gene family consists of C2C2 zinc finger domain and YABBY domain, which are relatively conserved among members. Phylogenetic relationship results classified the four sequences into four subfamilies; promoter and expression analyses showed that spinach YABBY genes were mainly expressed in leaves, especially in functional leaves. The expression of YABBY gene family members in spinach was affected by low temperature, salt stress and various hormone stress. These results provide a theoretical basis for cloning and functional analysis of YABBY gene, and provide theoretical support for understanding the molecular mechanism of spinach leaf development..
